Proteomic identification of Placental Protein 1 (PP1), PP8, and PP22 and characterization of their placental expression in healthy pregnancies and in preeclampsia

Introduction: Placental Protein 1 (PP1), PP8, and pp22 isolated from the placenta. Here, we aim to identify the PP1, PP8, and pp22 and protein expression patterns placenta and trophoblast them to reveal the involvement of the potential complications of pregnancy.

Methods: We analyzed PP1, PP8, and protein pp22 by LC-MS. We compare the behavior of the placenta from PP1, PP8, and pp22 to be dominated placenta-expressed PP5 / TFPI-2. Scores of placenta-specificity resulting from microarray data. Trophoblasts isolated from a healthy placenta and distinguished; Total RNA was isolated and target microarray analysis.

We were assigned the placenta to the following groups: control premature, early-onset preeclampsia, early-onset preeclampsia with HELLP syndrome, the control period, and late-onset preeclampsia. After histopathology, used for construction placental tissue microarray, immunostaining with anti-PP1, anti-PP5, anti-PP8, or anti-pp22 antibody, and immunoscoring.

Results: PP1, PP8, and pp22 identified as ‘nicotinate-nucleotide pyrophosphorylase’, ‘serpin B6’ and ‘protein disulfide isomerase-‘, respectively. Genes encoding PP1, PP8, and pp22 not dominated placenta-expressed, in contrast to PP5. PP1, PP8, and pp22 mRNA expression levels did not increase during trophoblast differentiation, in contrast to PP5. PP1, PP8, and pp22 immunostaining was detected mainly in the trophoblast, while PP5 expression is restricted to the syncytiotrophoblast. The PP1 immunoscore higher in late-onset preeclampsia, while PP5 immunoscore higher in early-onset preeclampsia.

Discussion: PP1, PP8, and pp22 expressed mainly in trophoblast but do not have regulations or specific trophoblast function. Different dysregulation PP1 and PP5 expression in both late-onset preeclampsia or early-onset reflect different pathophysiological pathways within this subset of preeclampsia.

Overexpression of B7-H4 promotes renal cell carcinoma tumor growth by recruiting neutrophils associated with upregulation of CXCL8

Checkpoint molecule B7 family members immunity H4 (B7-H4) play a role similar to the programmed death ligand-1 in tumor immune evasion by regulating the immune response of T-cell-mediated. However, in addition to the role of T-cell immunity, B7-H4 also affect the biology of tumor cells by promoting tumor cell proliferation, metastasis and angiogenesis.

In order to explore the effects of B7-H4 on the tumor cell biology, it is necessary to investigate the gene expression profiles when B7-H4 expressed. In this study, cells transfected 786-O stably express B7-H4. A microarray technique was then used to screen B7-H4-related genes differentially expressed (degs) in cell B7-H4 / 786-O compared to the negative control (NC) / 786-O cells.

Protein expression of degs deregulated, including the cells of non-metastatic 5, family members NME / NM23 5 (NME5), membrane metalloendopeptidase (MME), the blood vessels of non-inflammatory molecule 1 (VNN1), matrix metalloproteinase (MMP) 7, tumor necrosis factor, CXC motif chemokine ligand (CXCL) 8, CXCL1 and CC motif chemokine ligand (CCL) 2, were investigated using western blotting.

Renal papillary renal cell carcinoma mRNA sequencing data obtained from The Cancer Genome Atlas revealed that chemokines, including CXCL1 / 2/3, CXCL8, MMP7 and CCL20, was positively correlated with the expression of B7-H4 gene. Furthermore, 59 clinical renal cell carcinoma tissue was collected and analyzed by immunohistochemical staining. The results showed a positive correlation with B7-H4 CCL20 and CXCL8, and validated degs identified in tumor cells.
