Development of a multiplex assay for antibody detection in serum against pathogens affecting ruminants

Many infectious diseases that affect livestock impose significant economic burdens, and in some cases also a threat to humans and is classified as a zoonosis. Some zoonotic diseases transmitted by vectors and, due to environmental factors and socio-ecconomic complex, the distribution of many pathogens is changing, with a growing number of which are found in countries that were previously unaffected.

Here, we develop multiplex assays, microarray based suspension, is able to detect specific antibodies for the five important pathogen of cattle (three of them zoonoses) that currently appears in a new geographical location: Rift Valley fever virus (RVFV), Crimean- Congo hemorrhagic fever virus (CCHFV), Schmallenberg virus (SBV) Bluetongue virus (BTV) and the bacterium Mycobacterium complex tuberculosis.Using Luminex platform, polystyrene microspheres coated with recombinant protein from each of the five pathogens. The mixture of microspheres used for the simultaneous detection of antibodies against five diseases in accordance affecting ruminants.

The following pane sera included in this study: 50 sera from sheep experimentally infected with RVFV, 74 sera from calves and sheep vaccinated with SBV, 26 sera of cattle vaccinated with Mycobacterium bovis, 30 field sera from ruminant species differ infected CCHFV and 73 calf sera infected with BTV. Finally, to determine the diagnostic specificity of 220 field sera Spain frees her from the farm of five disease assessed. All sera were classified using a commercial ELISA specific to each disease, which is used in this study as a reference technique.

The results showed a multiplex test exhibited excellent performance characteristics with sensitivity ranging from 93-100% and specificity ranging 96-99% depending on the pathogen. This new tool enables the simultaneous detection of antibodies against five key pathogens, reducing the required sample volume and timing analysis where patogens is usually tested individually.

optoelectrical highly sensitive biosensors for diagnosis allergic multiplex

Compact multiplexing biosensor system has great potential for the diagnosis of diseases in which the detection of multiple biomarkers is required. Immunoglobulin E-mediated hypersensitivity syndrome is a primary immunodeficiency disorders associated with sensitization to allergens. Assessing immunoglobulin E (IgE) sensitization to allergens is an important strategy for the diagnosis of allergies.

Here, we report for the first time a reliable, flexible optoelectrical biosensor system and cost-effective for the simultaneous determination of total and allergen-specific IgE and IgG antibodies using immunogold-silver method of signal amplification. This biosensor is built on the usual digital versatile disc (DVD) to immobilize a 12 panel of allergen extracts or purified protein in a microarray format, as a proof of concept.

The biosensor multiplexing indicates the limit of detection of 0.26 IU / mL (624 pg / mL) and 14 ng / mL for IgE and IgG antibodies, respectively. The system is successfully applied in a cohort of 127 human serum samples, showed good sensitivity (97.6%) and specificity (85.7%), and a very good area under the curve (AUC) values ​​found in 0.977 (confidence interval, CI 0.957 to 0.990) compared and validated with immunofluorescence assay clinical references, confirming the excellent correlation between the two techniques.
